Skippers of U-145 include Heinrich Driver until December 1940 who also commanded U-20, U-23 and U-371; Rudolf Franzius to October 1941 who also commanded U-438; Heinz Schomburg until November 1941 who also commanded U-561; Reimer Ziesmer until December 1942 who also commanded U-236 and U-591; Hübschen ‘til May 1944 who also commanded U-2501 & U-2542; Hübsch until November 1944 who also commanded U-78 and Görner to the end of the war.

U-145 operated as a Frontboot (combat boat) and was attached to the 1st U-Bootflottille until December 1940, then in August 1941 she was transferred to the 22nd U-Bootflottille as a Schulboot (training boat). She was surrendered in May 1945, and was transferred to the British on 30 June 1945.

U-145 attacked no shipping during the war.
