The first Skipper of U-134 was Kapitänleutnant Rudolf Schendel who also served on U-19. On 20 January, 1943 command passed to Oberleutnant zur See (later Kapitänleutnant) Hans-Günther Brosin, who had formerly commanded U-634 from August 1942 until February of 1943.

U-134 was attached to the 3rd U-BootFlotille based at La Rochelle and took three war patrols in the Arctic against Allied convoys to Russia over December 1941 to February 1942. They attacked a German convoy in error, sinking one ship. Then U-134 operated in the North Atlantic for several patrols, another patrol off Florida where U-134 became the only U-Boat to shoot down a US Navy BLIMP. The BLIMP K-74 was shot down off Cuba on 19 July, 1943. U-134 had operations off Florida and in the Gulf of Mexico took place in July, 1942. U-134 operated in the Central Atlantic over November and December of 1942 and on 15 January, 1943 they lost one crewman in a non-combat situation. While returning to base from her Florida/Caribbean/Gulf of Mexico patrol, U-134 was lost with all hands.
