The first Skipper of U-133 was Kapitänleutnant Hermann Hesse who later commanded U-194 where he was killed. In February 1942, command passed to Oberleutnant zur See Eberhard Mohr who had come from U-148; he was lost with the boat.

U-133 was assigned to the 7th U-BootFlotilla based at St. Nazaire, France. She had one patrol in the North Atlantic then a successful break-through of the Straits of Gibraltar into the Mediterranean over the night of 21-22 December, 1941. On 14 March, 1942 the boat struck a mine outside Salamis Harbor and it is believed to have been a German mine - possibly one of her own.
